Phụ lục 1: Môi trường SC
Cho 1 lít môi trường
Thành phần hấp Thành phần lọc
Cornmeal = 15 g Bovine serum albumine = 2 g
Agar = 15 g Glucose = 0,5 g
Soytone = 8 g Cystein = 0,5 g
K2HPO4 = 1 g H2O = 200 ml
KH2PO4 = 1 g
MgSO4.7H2O = 0,2 g
Bovine hemine chloride = 15 mg
H2O = 800 ml
Hòa tan các thành phần hấp trong nước và điều chỉnh pH môi trường về giá trị
pH = 6,6 với NaOH 1N hoặc HCl 1N. Hòa tan các thành phần lọc trong nước cất vô
trùng.
Hấp tiệt trùng môi trường, để nguội đến khoảng 55 C; sau đó, cho các thành
phần lọc và nalidixic acid 10 g/ml vào qua màng lọc vô trùng (Davis và ctv.,
1980).
Phụ lục 2: Môi trường MSC
Cho 1 lít môi trường
Thành phần hấp Thành phần lọc
Corn meal agar = 17 g Glucose = 2 g
Bacto – agar = 4 g Cystein = 0,5 g
Bacto – peptone = 8 g Bovine serum albumine = 2 g
Bovine hemine chloride = 30 ml H2O = 100 ml
(Dung dịch 0,1% trong NaOH 0,05N)
MgSO4. 7H2O = 0,2 g
K2HPO4 (0,1 M) = 13 ml
KH2PO4 (0,1 M) = 87 ml
H2O = 800 ml
Hòa tan các thành phần hấp trong nước và điều chỉnh pH môi trường về giá trị
pH = 7,5 với NaOH 5N. Các bước tiếp theo được tiến hành tương tự như cách chuẩn
bị môi trường SC (Brumbley và ctv, 2006).

Cho 1 lít môi trường
Thành phần hấp Thành phần lọc
Bacto – Soytone = 8 g Glucose = 2 g
Hemin chloride = 15 ml Cystein = 0,5 g
(Dung dịch 0,1% trong NaOH 0,05 N) Bovine serum albumine = 2 g
MgSO4. 7H2O = 0,2 g H2O = 100 ml
K2HPO4 = 0,35 g
KH2PO4 = 1,1 g
H2O = 885 ml
Hòa tan các thành phần hấp trong nước và điều chỉnh pH môi trường về giá trị
pH = 6,6 với NaOH 5N. Hòa tan các thành phần lọc trong nước; sau đó cho vào môi
trường hấp đã nguội bằng một màng lọc vô trùng (Brumbley và ctv., 2006).
Phụ lục 4: Môi trường Phenol Red Carbohyrate Broth (Trần Linh Thước, 2003)
Cho 1 lít môi trường
Trypticase hoặc proteone peptone No. 3 10 g
NaCl 5g
Beef extract 1g
Phenol red 0,018 g
Nước cất 1 lít
Carbohydrate
: tùy nguồn carbon cần kiểm định ( manitol, sorbitol,...), thường được sử dụng ở nồng độ 1%
